Description Purpose
The purpose of this position is to work with the Principal Investigator (PI) and lab members on research aimed at understanding the structure and function of RNA molecular machines. The incumbent will use molecular biology methods to produce RNA and protein expression plasmids. They will transcribe and purify large structured RNAs by in vitro transcription, purify proteins by chromatography, and screen/optimize crystallization conditions. They will follow established workplace procedures to maintain an RNase free environment. The incumbent will also assist other laboratory staff, ensure regulatory compliance, and perform day-to-day maintenance of the laboratory. The incumbent will assist with generation of data and development of manuscripts for publication.

Duties & Responsibilities
Responsibilities are functional in nature, and performed under supervision. Specific tasks include, but are not limited to:
1) Perform, manage and assist with independent laboratory experiments carried out in the Principal Investigator’s (PI) laboratory that are essential for the achieving goals of the research projects. Prior experience is necessary in: DNA RNA isolation, DNA cloning, in vitro transcription, and protein purification. The candidate must be able to modify and optimize research protocols as needed for the successful completion of the specific experiment.
2) Screen candidate RNA and RNP complexes for crystallization, and optimize crystal growth conditions.
3) Function in the capacity of laboratory manager. This person will be responsible for general oversight and compliance of students working in the laboratory on a day-to-day basis. Conduct daily laboratory maintenance including ordering / purchasing of supplies and equipment, and keep proper records.
4) Make reagents / solutions for experiments; assist in maintaining laboratory glassware and sterilization procedures.
5) Maintain a well-organized notebook documenting experimental protocols and data. Consult at regular intervals with the PI for guidance in experimental design and for progress reports.
6) Informally supervise and train other staff and students.
7) Communicate with others in the lab and the department on ongoing experiments, shared resources, maintenance and safety issues.
8) Ensure compliance with safety regulation

Supervisory/Lead Role
This position reports to the Principal Investigator.

Physical Demands
Activities require manual skill to perform accurate micropipetting of samples, mixing of small volumes of DNA/RNA, standing/sitting by the bench, lifting of fluid-containing carboys, bending, carrying lightweight objects (up to 40 lbs), and walking back and forth from animal facility for transporting mice, which can be a medium distance, traversing stairs or using elevators. Good hand-eye-coordination, precision of movements, and attention to detail is required.

Work Environment
Work will be performed in a standard laboratory environment including using computers and laboratory equipment with electrical connections. Some work will be performed in the cold room and some laboratory chemicals have an odor or toxic fumes. Some work will be performed in the dark room either under dim red light or in the complete darkness using night vision equipment. There will be exposure to murine eye tissues, laboratory animals, and radiation, however protective measures will be provided. Responsible to know and comply with all safety regulations.

The following has been determined as a possible job exposure of this position requiring WVU Environmental Health and Safety medical monitoring:
1) Contacting Hazardous Chemicals – Daily or weekly, using strong acids or caustics, fuel or tar products or cancer causing chemicals
2) Hazardous Materials Responder- for those certified as OSHA Hazardous Materials Responders or lab employees that clean chemical spills in their work
3) Scientific Research- research with nanoparticles, lasers, anesthetic, fuels or mining, aquatic or diving etc.

Requirements Requisite Qualifications
Education and/or Experience
1) Bachelor’s degree in biochemistry/molecular biology or related field
2) 6 months – 4 years of experience in biological research
3) Equivalent combination of education and experience

1) Knowledge and laboratory experience in general molecular biology techniques.
2) Skilled in DNA/RNA isolation and Polymerase Chain Reaction (PCR).
3) Expression and purification of recombinant proteins from E. coli
4) Knowledge of crystallography techniques.
5) Knowledge of laboratory environment and appropriate procedures.
6) Ability to prepare reagents.
7) Ability to present scientific data both orally and visually.
8) Knowledge of Microsoft Office suite.
9) Skilled at using computers and applicable programs/applications.
10) Skilled at assessing chemical concentrations/amounts of various chemical
components in solutions.
11) Skilled at using scientific rules and methods to solve problems.
12) Skilled at using related laboratory equipment.
13) Good organization skills and self motivation.
14) Willingness and ability to learn new protocols/ procedures in a rapid manner.
15) Ability to get along with others in a group setting.
